Margarete Henriette Katzenstein was born in 1905 in Frankfurt am Main, where her father was a prominent physician. She studied at the Universities of Frankfurt and Heidelberg. In 1929 she became an assistant at the Medizinische Universitaetsklinik, a position which she had to leave in 1933. She then began working at the Krankenhaus der Israelitischen Gemeinde, where she became the chief assistant to Professor Simon Isaac.
